Syllabus of lectures

1-4. Pojem struktury makromolekul, základní strukturní motivy proteinů a nukleových kyselin, struktura sacharidů a membrán.
5. Výpočetní metody, molekulová mechanika a dynamika, simulované žíhání.
6. Příprava vzorku, sekvenace nukleových kyselin, proteinů a sacharidů.
7. Optické metody charakterizace biomakromolekul: cirkulární dichroismus, infračervená spektroskopie.
8-9. Rentgenová strukturní analýza. Příprava krystalů, difrakční experiment, metody řešení fázového problému, mapy elektronové hustoty, výstavba strukturního modelu.
10. Elektronová mikroskopie.
11-12. Nukleární magnetická rezonance. Izotopové značení, NMR experiment, přiřazení frekvencí ve spektrech, určení geometrie (NOE, interakční konstanty), dynamika proteinů.
